Health & Medicine Week May 19, 2008 Current study results from the report, ‘Temsirolimus downregulates p21 without altering cyclin D1 expression and induces autophagy and synergizes with vorinostat in mantle cell lymphoma,’ have been published. “To investigate the mechanisms of antiproliferative effect induced by the mammalian target of rapamycin (mTOR) inhibitor temsirolimus in mantle cell lymphoma (MCL). The antiproliferative effect of temsirolimus on three well-defined MCL cell lines was examined by the MTS assay,” researchers in the United States report (see also Mantle Cell Lymphoma Therapy). website mantle cell lymphoma
“Induction of cell-cycle arrest, autophagy, and apoptosis were determined by fluorescence-activated cell sorting analysis. The molecular mechanisms underlining these effects were determined by Western blot. Synergy between temsirolimus and vorinostat were examined by MTS assay and the combination index was calculated. Temsirolimus has antiproliferative activity in three MCL cell lines in a dose-and time-dependent manner. Mechanistically, temsirolimus inhibited mTOR, as evidenced by inhibition of ribosomal S6 phosphorylation, and induced cell-cycle arrest in the G(0)/G(1) phase and a decrease in p21 expression without altering p27 or cyclin D1 levels. Furthermore, temsirolimus increased the number of acidic vesicular organelles and the amount of microtubule-associated protein 1 light-chain 3 processing, which are characteristic of autophagy, without induction of apoptosis. These changes were not associated with alteration in phosphorylated extracellular signal-regulated kinase (ERK), beclin-1, Bax, or Bak levels. In contrast, treatment of these cell lines with the histone deacetylase inhibitor vorinostat decreased ERK phosphorylation, activated caspase 3, and induced apoptosis. Moreover, temsirolimus synergized with submaximal concentrations of vorinostat in all MCL cell lines. This is the first report of temsirolimus-induced autophagy in MCL, and of vorinostat inhibition of ERK phosphorylation in MCL,” wrote V.Y. Yazbeck and colleagues, University of Texas, M. D. Anderson Cancer Center.
The researchers concluded: “Collectively, these data suggest that the combination of temsirolimus and vorinostat have synergistic antiproliferative activity in MCL cells by distinctively targeting apoptosis and autophagy.” Yazbeck and colleagues published their study in Experimental Hematology (Temsirolimus downregulates p21 without altering cyclin D1 expression and induces autophagy and synergizes with vorinostat in mantle cell lymphoma. I think Musharraf is better than looter: BB & NS. Since he does not belong to Jagirdar class, therefore many people are not very comfortable with him. He is one of the best leaders we had in 60 years. No party has ever given any Economic and Business Development Plan for Pakistan. rather these politicians have looted the country. People can not live in USA or EU with working for two months even, how-come BB and Sharifs live many years very lavishly. This is looted wealth from poor Pakistanis. Can CJ or SC can take this notice?????
I think Musharaff is a great leader……..We need to realize that every country has its dark realities and pakistan is no exception….Being en expat i would like to say that musharaff’s forign policies are the best and our country badly needs him. Just imagine if musharaff is gone and benazir takes over what would happen?? we dont want money launderers ruling our country ones again or do we?? and please people be REAL and for god’s sake stop calling saddam and musaraff buddies!!! there is a HUGE difference between the pakistan before mushraff’s rule and the pakistan today!! One thing that i wanted to say but could’nt anywhere was that it is very very amusing to see people like nawaz sharif offering their condolences to media on ban of some progs!!!What the heck…..i think he has forgotten his times when he did not let the media even speak against him and there was ony one GOVERNMENT OWNED channel which spoke the words of nawaz shariff only!!!
